Once you have the basic idea about the story in your mind, you can start by building the characters and then go on knitting the story around their lives, stating the obstacles suffered by them and the problems handled and so on. Thus you could start by building the characters and then go on to the story. Think from a multidimensional perspective and look for various directions that the plot can be built in.
